Promyelocytic leukemia (PML) nuclear bodies (NB) facilitate the proteasomal degradation of protein aggregates through the SUMO-targeted ubiquitin ligase pathway. A study now reveals a non-proteolytic role for PML-NBs in preventing TDP-43 aggregation that still relies on the SUMOylation–ubiquitylation cascade but uses it to direct protein aggregates towards p97 ATPase-mediated disaggregation.Nature Chemical Biology, Published online: 30 June 2025; doi:10.1038/s41589-025-01945-w
Legionella effector LnaB, a recently identified actin-activated AMPylase, mediates phosphoryl-AMPylation in a yet-unknown manner. Here the authors unveil a two-step catalytic process involving dual ATPase–AMPylase activity based on structural studies, providing a biochemical basis for AMPylation.Nature Chemical Biology, Published online: 27 June 2025; doi:10.1038/s41589-025-01952-x
Dysregulated R-loops can compromise genome stability, leading to pathological consequences. Li et al. demonstrate that R-loop-induced poly(ADP-ribose) polymerase 1 activation orchestrates exonuclease 3′–5′ domain-containing 2-mediated R-loop resolution, thereby safeguarding genomic integrity.Nature Chemical Biology, Published online: 27 June 2025; doi:10.1038/s41589-025-01950-z
The development of antibody–drug conjugates remains a challenge in part due to the lack of three-dimensional structural information that must account for the inherent flexibility of antibodies and drug payloads. This Perspective discusses computational methods to guide the design of antibody–drug conjugates.Nature Chemical Biology, Published online: 26 June 2025; doi:10.1038/s41589-025-01976-3

The discovery of seven soybean genes completes the biosynthetic pathway to major glyceollins, granting access to heterologous production and exploration of their antimicrobial potential.Nature Chemical Biology, Published online: 26 June 2025; doi:10.1038/s41589-025-01957-6
Description of the cryo-EM structures of the parathyroid hormone type 1 receptor associated with Gq reveals two distinct extracellular conformations with N-glycans stabilizing a ligand-tilted conformation.Nature Chemical Biology, Published online: 25 June 2025; doi:10.1038/s41589-025-01943-y
Despite the importance of indole in plant defense and communication, its biosynthesis in core eudicots remains elusive. Here the authors report the identification of TSB-like, a pseudoenzyme that hijacks an enzyme of tryptophan biosynthesis to produce indole. The library can offer a variety of group and personal assistance to you.
